1998 年に設立された「ゲノム創薬フォーラム」の趣意書には、ゲノム創薬を以下のように定義していた。

「医薬品を作り出す創薬という作業は、これまで一定の目的を定め、多数の化合物や天然物をスクリーニングすることで行われてきた。例えば、新しい受容体を誰かが発見すれば、それを創薬研究者たちは競って標的とした。現在も有効なこのアプローチを続けるうち、優れた医薬品を発見するためには、まず標的自身が新しく、ユニークでなければならないことを私たちは知った。これを可能にする創薬における画期的変化がゲノム科学をベースにした標的の発見とそのダイナミックな活用であった。これをゲノム創薬研究プロセス(Genomic Drug Discovery Process) と呼ぼう。」 20 年前にこのように定義されたゲノム創薬プロセスは、現在多くの成功例を生んでいる。

2017 年の世界市場でのTop100 の医薬品において、日本発の製品は12 品目(低分子10、バイオ2)であり米国に次いで世界2 位の地位にある。現在の成功を未来への更なる成功として継続発展させていく為にも、産学からの研究者が最新の仮説とデータを基に議論する場を提供することは貴重である。特に、プレコンペティション段階の創薬研究を議論する場として、参加者の多様な経験・知識に基づき自由闊達で多様な意見を引き出し、日本発の新しいゲノム創薬のアイデアを創発する場を提供したいと考えている。

ゲノム創薬の目指すものが、従来の治療薬のみならず、予防・未病へと進み、その診断や予測にBig Data やAI が使われ、テクノロジーの進展はこれまで以上に加速している。このような状況のなかで『ゲノム創薬・創発フォーラム』は、創薬研究プロセスの創薬コンセプトの形成、ターゲット探索、モダリティー探索において、何をどのようになすべきかを産学ともに考え、最新のゲノム科学・情報科学を取り込んだ創薬研究を強力に支援・推進するためのシステム作り、ひいては日本におけるゲノム創薬の成功と発展に寄与することを目標としたい。企業、アカデミアからの多数の参加を期待する。

ゲノム創薬・創発フォーラム設立の背景
「ゲノム創薬フォーラム」は、野口照久代表および新井賢一副代表を中心に1998 年に設立された。“Human genome project”を中心として急速に進歩しつつあった網羅的な遺伝子発現プロファイル解析、疾患に関わる遺伝子多型の同定、タンパク質の発現と立体構造を解析するプロテオミクスなど、日本のみならず世界の最先端研究を紹介し、その成果を如何に創薬に取り込むかを産学からの参加者で共に議論してきた。

2013 年には新井賢一を代表として「ゲノム創薬・医療フォーラム」と改名し、網羅的なゲノム解析による横の広がりと共に、疾患の治療(医療)という縦の広がりを加えた。がん治療を例にとってみれば、薬剤の標的としてどの分子が最も適切であるのか、制がん剤の副作用がどんなメカニズムで起こるのか、効く患者と効かない患者の差がどこにあるのか等、これまで以上に医療の目標を明確にしていく必要があり、こうした視点から臨床開発企業や医療機器に関与する企業、病院・医療関係者の参画も募り議論の輪を広げることを企画し、成果を挙げてきた。しかし、2018 年には新井賢一が急逝したことを受け、「ゲノム創薬・医療フォーラム」の解散が同年11 月に決議され、本活動は終息することとなった。

しかしながら、産学からの参加者が自由な雰囲気で最先端の科学を議論する場を提供するフォーラムの存在意義は依然大きい。また、ゲノム科学と情報科学の統合がイノベーション創出の鍵となっており、情報科学の専門性を持つアカデミア・企業の専門家の参画を募り議論する場を提供することが社会と時代の要請である。

そこで我々は、これまでの体制を一新し、2019 年4 月を目途に、新たに「ゲノム創薬・創発フォーラム」を設立することとした。「創発」とは部分の性質の単純な総和にとどまらない性質が全体として現れることを意味し、異なる分野の専門家が議論することにより所定の意図を超えたイノベーションが誘発されることを期待して会の名称に取り入れた。

本会は、シンポジウムを年3回開催する。企業の創薬研究者とアカデミアの研究者による積極的なコミュニケーションによってテーマを選定する。革新的創薬に繋がる可能性を秘めた日本発の先見的な研究成果を紹介する機会をつくり、産学からの参加者の議論と交流の場を提供し、明日の創薬に活かしたい。そのためには、フォーラムの計画と運営に、産学の方々の協力が必須となる。

代表は松島綱治が務め、幹事を刷新する。中心とする疾患領域を、がん、炎症・免疫難病とし、一層活発なフォーラムとして活動していきたい。 [/su_tab]

Genome-based drug discovery was defined in the founding statement of the Genome-based Drug Discovery Forum, which was founded in 1998.
“The process of drug discovery has been driven by setting defined goals and then screening a lot of chemicals and natural products. For instance, when a novel receptor was discovered, drug discovery researchers were competitively targeted. Repetitive application of this approach, which is still effective today, has revealed that careful target selection is a key first step in the process. The target should be novel with unique properties to support discovery of an excellent new drug. Outstanding genome science-based progress has made it possible to discover novel targets and incorporate them into dynamic drug discovery efforts. Let us call this the genome drug discovery process.” We now can see many successful examples of genome drug discovery as defined 20 years ago.

In the global pharmaceutical drug market in 2017, 12 of the top 100 new drugs originated from Japan, which is the 2nd highest contributor among the listed countries. To sustainably increase current success levels in the future, it is important to offer opportunities for researchers in both academia and industry to openly discuss the most up to date hypotheses and research data. In particular, at noncompetitive stages of drug discovery, we want to offer opportunities to induce innovative emergence (So-hatsu) of novel ideas in genome based drug discovery across Japan, whereby fruitful and diverse opinions are evoked from participants’ broad experience and deep knowledge.

The scope of genome based drug discovery includes protection from and prevention of diseases beyond the discovery of conventional therapeutics. Big data and AI are used for diagnosis and forecasting, and technological progress is accelerating more than ever. Under these circumstances, the goal of the genome drug discovery and innovation forum is to contribute to the success and advancement of genome drug discovery in Japan by fostering discussions between academia and industry regarding what novel directions to pursue, how to implement conceptual drug discovery processes, and selection of target molecules and drug modalities. We urge the participation of both academia and industry.

History of the genome drug discovery and emergence forum

The Genome Drug Discovery Forum was founded by Dr. Teruhisa Noguchi (President) and Dr. Kenichi Arai (Vice President) in 1998. Much recent research has involved global analysis of genome expression profiling, identification of disease-related genomic polymorphisms, and/or proteomics analysis of protein expression and 3D structure. The first forum involved participants from academia and industry in a discussion of how to incorporate those outcomes into drug discovery.

In 2013, Dr. Kenichi Arai broadened the forum’s activity not only horizontally in global genomic analysis, but also vertically into therapeutic fields. For example, we needed clearer strategies in cancer therapeutic research, which required identification of the most suitable molecular targets, a better understanding of the mechanism(s) of adverse drug effects, and the ability to diagnostically distinguish responders from non-responders. To this end, the forum extended its discussion network to incorporate drug companies conducting clinical drug development, and medical device development companies, hospitals, and medical workers. The forum temporarily terminated these activities in November 2018 after Dr. Arai passed away.

Nonetheless, the provision of forums where participants from academia and industry could get together to discuss the most advanced science in an open atmosphere can have a significant positive impact on innovation. Currently, the integration of genome science and information science is becoming critical. Now is an opportune time to offer in demand discussion opportunities by inviting information science experts in academia and industry.

Therefore, we organized a new consortium, the Genome Drug Discovery & Emergence (so-hatsu) Forum, which renews most aspects of the preexisting forum in April 2019. So-hatsu refers to the overall result exceeding the expected outcome. Applied to the new forum, “Emergence” conveys the expectation of synergy from bringing together innovative experts from many fields from both academia and industry.

The forum organizes scientific symposia three times a year. We select each theme through active communication with drug researchers in industry and medical researchers in academia. We create opportunities to share promising research outcomes that may lead to innovative drug discoveries. We also urge discussion and interaction among participants in academia and industry to support drug discovery tomorrow. The support and cooperation of academia and industry are critical.

Dr. Kouji Matsushima serves as president and represents the board members. The major disease areas the forum focuses on include cancer, inflammation, and immunological disorders, and we hope to steadily expand our areas of activity.[/su_tab]
